Mold identification using MALDI-TOF mass spectrometry, particularly with systems like VITEK MS, offers a rapid and reliable method for identifying fungal species, including molds, in clinical microbiology laboratories. 

Matrix-assisted laser desorption ionization time-of-flight (MALDI-TOF) mass spectrometry is a technique that uses a laser to ionize and desorb molecules, allowing for their mass analysis, which is then compared to a database for identification. 

Procedure for mold identification

  • Sample Preparation: Mold samples are prepared, often through extraction, to obtain a suitable protein profile for analysis.
  • Spectra Acquisition: The prepared sample is placed on a MALDI target plate, and a laser beam is used to ionize the proteins, creating a mass spectrum.
  • Database Comparison: The generated mass spectrum is compared to a database containing spectra of known fungal species, allowing for identification.

VITEK MS: The VITEK MS system is a commercial MALDI-TOF MS platform used for microbial identification, including fungi, and is widely used in clinical laboratories.

  • Examples of Mold Species Identified MALDI-TOF MS has been used to identify various clinically relevant fungal species, including Aspergillus spp., Fusarium spp., Scedosporium spp., and Mucorales.
