LAMP and PCR: Introduction, Differences, and Keynotes

Introduction

LAMP stands for Loop-mediated Isothermal Amplification while PCR full form is polymerase chain reaction and LAMP is the nucleic acid amplification assay. The lamp is a newer technique whereas PCR is older. The LAMP is comparatively more efficient and cost-effective than PCR testing.

Differences between LAMP and PCR Assays

Differences between them are as follows-

LAMPPCR
Emerging MethodOlder method
Exponential accumulation of additional dsDNA is faster.Slower
No need for multiple temperature cyclesMultiple temperature cycles are needed.
LAMP amplification also depends on the low range of heat at around 60-65ºC.A higher range of heat is required i.e. 56-95ºC.
Result within minutes (average 30 minutes)Result within hours (average 4 hours)
It can ve handled in a simple setup. It needs an advanced level of setup.
Low manpower and basic training are sufficient.High manpower and highly skilled laboratory personnel are mandatory.
Equipment name-MicrosensDx RapiPRO™ Loop-mediated Isothermal Amplification (LAMP) SARS-CoV-2 testBioRad CFX-96, Applied Biosystem-Qunnstudio 5, RotorGene Q
It has far greater accuracy and sensitivity than antigen testing for SARS-CoV-2.It has too.
More efficient and cost-effective Costly
RapiPRO™ LAMP for SARS-CoV-2 test has widely applied on large industrial sites and in airports. It is hard to install in such areas.
No need for expensive reagents or instrumentsIt needs expensive reagents or instruments.
Highly sensitive (nearly 100-fold more sensitive than conventional RT-PCR methods).Comparatively less sensitive

Keynotes

  1. Loop-mediated isothermal amplification (LAMP) assay is a rapid diagnostic for COVID-19.
  2. RT-PCR for COVID-19 is a gold standard assay.
  3. Lamp is a nucleic acid amplifixation assay and higly senstive than conventional RT-PCR methods.
  4. PCR is a nucleic acid targets assay i.e. N2 (nucleocapsid), E (envelope), S (spike protein),etc.
